Projekte zum Thema Saccharosetransport

  • Bestätigung von Protein-Protein-Interaktionen der Saccharosetransporter (SUTs) mittels Split Ubiquitin System, Bimolecular Fluorescence Complementation, FRET acceptor bleaching etc.
  • Untersuchung der physiologischen Funktion von SUT-interagierender Proteinen hinsichtlich ROS Detoxifikation und abiotischer Stresstoleranz.
  • Untersuchungen zum Makromolekültransport über Plasmodesmen unter Verwendung transgener Pflanzen
  • Untersuchung der lateralen Mobilität von Membranproteinen in Mikrodomänen über Fluorescence recovery after photobleaching (FRAP).
  • Identifizierung von regulatorischen Aminosäuren von Saccharosetransportern durch gezielte PCR-basierte Mutagenese und Untersuchung von Dimerisierung, targeting, Phosphorylierungsverhalten und pH-Abhängigkeit der Mutanten.
  • Aufklärung des Zusammenhangs zwischen dem Saccharosestoffwechsel und Brassinosteroid Signaling anhand transgener Pflanzen, in denen die Expression des Saccharosetransporters SUT2 bzw. BR-Biosynthese und BR-Signaling beeinträchtigt sind.
  • Untersuchung der veränderter Mykorrhizierungseffizienz transgener Tomatenpflanzen mit reduzierter Expression SUT-interagierender Proteine
  • Rolle des Saccharosetransporters StSUT4 für die Blühinduktion, Ethylenproduktion und Gibberellin Signaling. Bestätigung der bislang identifizierten SUT4-Interaktionspartner.
  • Identifizierung von Transkriptionsfaktoren, die an der Expressionskontrolle von Saccharosetransportern beteiligt sind über das Hefe-1-Hybrid-System, ChIP-Seq Analysen bzw. Transaktivierungsstudien.

Projects related to sucrose transport and partitioning

  • Confirmation of protein-protein interactions of plant sucrose transporters via Split Ubiquitin System, Bimolecular fluorescence complementation, FRET acceptor bleaching etc.
  • Investigation of the physiological function of SUT-interacting proteins with respect to ROS detoxification and abiotic stress tolerance.
  • Investigation of macromolecular transport via plasmodesmata by help of transgenic plants
  • Lateral mobility of membrane proteins in membrane microdomains determined by Fluorescence recovery after photobleaching (FRAP).
  • Identification of relevant amino acids within plant sucrose transporters for dimerization, targeting, phosphorylation and pH dependency by PCR-based site directed mutagenesis. Functional characterization in yeast and in plants.
  • Cross-talk between sucrose metabolism and brassinosteroid signaling by manipulation of expression of sucrose transporter SUT2 and members of BR signaling and biosynthesis.
  • Investigation of modified mycorrhization efficiency of transgenic tomato plants with altered expression of SUT-interacting proteins.
  • Role of StSUT4 for flower and tuber induction, ethylene production and GA signaling. Confirmation of SUT4-interacting proteins.

Identification and confirmation of transcription factors involved in the regulation of sucrose transporter expression by yeast-one hybrid studies, ChIP-Seq experiments and transactivation studies.
