Wir bieten jeweils aktuelle Themen aus den verschiedenen Forschungsvorhaben für Abschlussarbeiten von Bachelor- und Master-Studierenden.

Im Verlaufe der Bachelor- und Masterarbeit werden genetische, entomologische, chromatographische, fluoreszenzmikroskopische und weitere zellbiologische Analysen insbesondere von Mutanten und transgenen Pflanzen und Algen mit modifizierter Pigmentsynthese oder Stressreaktion, durchgeführt.

1. The green alga Chlamydomonas reinhardtii - an excellent tool to study control of photosynthesis and chlorophyll biosynthesis  

Chlamydomonas is a unicellular green algae species, which offers several advantages in studying tetrapyrrole biosynthesis, photosynthesis, and related processes. It is capable of heterotrophic growth, if provided with reduced carbon source. Therefore, defects in chlorophyll biosynthesis or photosynthesis, which normally lead to a lethal phenotype in plants, can be studied in algal mutants with the same genetic lesions. Unlike angiosperms, e.g. widely studied Arabidopsis thaliana or Nicotiana tabacum, C. reinhardtii also produces chlorophyll in the dark, due to the presence of light-independent protochlorophyllide oxidoreductase. This provides another advantage, because it is possible to study mutants impaired in tetrapyrrole biosynthesis, which display increased sensitivity to light. Based on the studies of C. reinhardtii, we are offering students BSc or MSc projects on the following aspects:

 

1.1 Analysis of non-characterized enzymatic steps in tetrapyrrole biosynthesis, including the identification of unknown enzymes and regulatory factors. Initial step of this study is the characterization of mutants affected in the tetrapyrrole metabolism or the posttranslational control of this pathway.

 

1.2 Identification of the regulatory proteins and signalling components involved in tetrapyrrole-mediated and 1O2-induced chloroplast retrograde signaling. Introductory step of this study is the initial characterization of signalling mutants, which have been obtained from previous mutant screens and are available in the lab

 

1.3 Identification and characterization of UV-mutagenized strains with interesting yellow, orange and brown pigmentation. These mutants are available in the lab and ready for physiological, biochemical and genetic analysis.

2. Regulation der Tetrapyrrolbiosynthese

Analyse von transgenen Pflanzen mit deregulierter Chlorophyll- oder Hämsynthese mittels molekulargenetischer und biochemischer Methoden.

 

2.1      In-vivo Charakterisierung der posttranslationalen Kontrolle der ALA Synthese mit Glutamyl-tRNA Reduktase (GluTR) und Glutamat-1-semialdehyd Aminotransferase (GSAAT).

Diese Arbeiten schließen die funktionelle Charakterisierung von Regulationsfaktoren dieser Enzyme ein, wie z.B. die Regulatoren FLU und GBP, das Chaperone cpSRP43, die Clp Protease, oder die Thioredoxine und NTRC für Redoxkontrolle. Aber auch nach weiteren Interaktionspartnern für die Kontrolle der ALA-Synthese wird gesucht.

2.2 Posttranslationale Kontrolle in dem Mg-Zweig der Tetrapyrrolbiosynthese. Diese Arbeiten befassen sich augenblicklich mit Analysen der Redoxkontrolle und der Chaperonfunktion von cpSRP43. Weitere posttranslationale Kontrollmechanismen werden mit neuen Regulatoren und Chaperonen, z.B. CPP1, TPR-Proteine, in der Zukunft untersucht.

3. Die Verknüpfung von Chlorophyllbiosynthese mit dem Einbau von Chlorophyll in die Photosynthese-Proteine in der Thylakoidmembran.

Die Arbeiten werden mit LHC-like Proteinen, z.B. LIL3 und OHPs, fortgesetzt. Außerdem befassen wir uns mit dem kürzlich identifizierten Recycling des Chlorophylls. Mittels Co-Immunpräzipitation, Affinitätschromatographie, Ultrazentrifugation, nativer Gelelektrophorese, und Massenspektrometrie werden die Interaktionen der Proteine für Chlorophyllbiosynthese und Integration des Chlorophylls in die Proteine der Photosynthese nachgewiesen.

Weitere Teilprojekte befassen sich auch mit weiteren Assemblierungsfaktoren und Chaperonen für die Organisation der Chlorophyllsynthese und der Biogenese der Thylakoidmembran.

4. Funktionsanalyse der beiden Isoformen der Ferrochelatase in der Hämsynthese

 

 

5. Lokalisation von Enzymen der Tetrapyrrolsynthese in Subkompartimenten der Plastiden

Diese Projekte tragen zum besseren Verständnis der Organisation der Chlorophyll- und Hämsynthese bei. Der Aufenthaltsort der einzelnen Isoformen von Enzymen innerhalb der Plastiden trägt zur Aufklärung ihrer Funktion bei. Damit wird geklärt, wie in einem verzweigten Stoffwechselweg Metabolite auf die Synthesewege verteilt werden: Chlorophyllsynthese in der Thylakoidmembran, Differenzierung der Hämsynthese für inner- und außerplastidäre Häm-abhängige Proteine.

6. Analyse des pflanzlichen Riboflavin (Vitamin B2)-Biosyntheseweg und die FAD/FMN Synthese in Plastiden und Zytoplasma

Analyse von Mutanten in der Riboflavin- und Flavinsynthese. Identifizierung von Genen für neue Enzymen der Flavinsynthese und für Transporter des Riboflavins und der Flavine.

Biochemische, genetische und massenspektrometrische Methoden werden eingesetzt, um die Regulation der Stoffwechselwege zum FAD und FMN in Plastiden und Zytoplasma zu unterscheiden und die Auswirkungen deregulierter Flavinsynthese auf die Funktion von FAD- bzw. FMN-abhängigen Enzyme in allen zellulären Kompartimenten zu analysieren.
