A Masters Project Studentship is available in the research group of Prof. Peter Hegemann in the Institute for Biology, Experimental Biophysics, Humboldt University of Berlin. This is an exciting opportunity to work on a project involving the expression and purification of Enzymerhodopsins that show light-regulated enzyme activity. The cyclic nucleotides cAMP and cGMP are ubiquitous second messengers, known for regulating several crucial cellular processes, including, basic metabolism, gene expression, differentiation, and cell survival. In this project, we shall characterize the photophysical and structural properties of the rhodopsin-guanylyl cyclase (RhGC) that produces cGMP in response to green light, from either Catenaria anguillulae or Blastocladiella emersonii. Specifically, we wish to study the structure of these proteins using cryo-EM technique. The structure and properties of the corresponding adenylyl cyclases (CaRhACs) will also be studied as these photoreceptors proteins have great potential for application in optogenetics and possess multiple advantages over the currently existing tools to manipulate cyclic-nucleotide dynamics in living cells. The candidate should have a Bachelors degree in Biophysics, Biochemistry, Biology or a related area. Experience in protein expression and purification is advangage but not essencial. Research Area:
Molecular characterization and improvement of bimodal switchable Opto-GPCRs used for the activation of G-alpha signaling in central neurons.
The project will includer structure-based design, medium size screening methods, electrophysiology, biochemistry, spectroscopy and high-resolution imaging.
Qualification profile:
We aim at attracting the best talent in the respective research fields and look for:
• an outstanding university degree (master/diploma or equivalent) and Dr. or PhD degrees in biochemistry, chemistry, physics, biology, or related field of physical sciences
• Dr. or PhD degree from an excellent research group / University
